Abstract

The ability to understand the principle of Photometry which utilizes the absorption of light to obtain concentration of a sample in question in this particular experiment Lowry method was used to determine concentration of protein by using colorimetric technique . This was achieved by plotting the standard curve to determine the unknown according to (Stoscheck 1990 . The graphical representation gives a linear relationship which obey the Beer-Lambert

Law but the interaction of the reagents may affect the absorbance giving incorrect reading but previous procedures have been invented as a modification of Lowry method which are not influenced by pH , salts and the turbidity of the sample (Penkova et al , 1996 , the results were evidence of precision and accuracy

Introduction

The discovery of spectrophotometer has made it possible for the quantification of biological macromolecules . Photometry principle is applied by measuring the amount of light absorbed by the sample in question according to (Blauch 2009 . spectrometer operate by measuring amount of light that is able to pass through which is reflected on the detector , to accomplish the task a suitable wave length of 540 nm is selected which can be detected by the human eye on the measuring device
